Background
Purinergic signaling involves cell signaling pathways that are activated by nucleosides and nucleotides via specific purinergic receptors (1). Purinergic receptors include two large families, P1 receptors that are activated by nucleosides and P2 receptors that are activated by nucleotides. P2 receptors are further divided into ionotropic P2X receptors and metabotropic P2Y receptors. P2Y purinoceptor 12 (P2Y12) is a member of the P2Y receptor family (2). As G protein-coupled receptors, P2Y12 are activated by ADP and ATP to drive downstream second messenger pathways. P2Y12 is required for normal platelet aggregation and blood coagulation (3). In addition to their role in thrombosis, P2Y12 receptors drive purinergic signaling in the central nervous system (CNS). P2Y12 receptors are enriched in microglia and regulate nucleotide-dependent microglia chemotaxis and morphology (4). Furthermore, P2Y12 receptors regulate specialized microglia/neuron junctions that provide microglia-dependent protection of neuronal functions in response to brain injury and disease (5).
