Background
ATP-binding cassette sub-family D member 3 (ABCD3) is a member of the ABC transporter family, a group of evolutionarily conserved membrane proteins that use ATP hydrolysis to transport diverse substrates across biological membranes (1). ABCD3 was first cloned from rat liver peroxisomes as a 70 kDa peroxisomal membrane protein named PMP70 (2). At the peroxisomal membrane, ABCD3 utilizes a nucleotide-binding domain (NBD) and a transmembrane domain (TMD) to create a cavity that enables transport of its substrates, fatty acyl-CoAs, from the cytosol into the peroxisome for β-oxidation (3). Mutations in the gene have been linked to hepatosplenomegaly (4).
